The invention discloses a CYP2C19 gene detecting method which comprises the following steps: extracting sample DNA; designing a specific primer for amplifying a CYP2C19 gene segment; utilizing singletube multi-PCR amplification reaction to obtain a target segment; designing a single base extension primer for the CYP2C19 gene segment; extending the single base of the CYP2C19 gene segment; and analyzing a nucleic acid mass spectrum for measuring a target DNA sequence. The CYP2C19 gene detecting method can effectively perform CYP2C19 gene polymorphic screening on a patient, and lays clinical andscientific research basis for performing personalized accurate diagnosis and treatment.
