The assay method includes the following steps that a tracer area is determined and a ground map of the tracer area is drawn; the type of the tracer agent is determined; the delivery quantity of the tracer agent is determined; a delivery well and monitoring wells are selected and distributed; delivery and sampling of the tracer agent are carried out; detection of a tracer sample is carried out; a result of the groundwater migration path is summarized; and result interpretation of the groundwatermigration path is carried out. According to the assay method of the migration path of the groundwater by using the tracer agent, an assay method of a sodium fluorescein tracer agent is adopted, a time-concentration relation curve is drawn, according to the time-concentration relation curve and the straight line distance between the delivery well and the monitoring wells, the groundwater average velocity and the groundwater flow direction of the delivery well and the monitoring wells are measured, then hydraulic power relation conditions of the groundwater in and out of the tracer area are found out, the groundwater flow rate is determined, and the main direction of the groundwater flow direction is determined.
