Problems solved by technology

Therefore, AFP is still not ideal as a marker for the diagnosis of liver cancer. The diagnosis of liver cancer based on AFP alone is likely to cause missed diagnosis and misdiagnosis. It is urgent to develop more simple, sensitive and reliable methods to improve the detection rate of early liver cancer.

Embodiment Construction

[0033] The solution of the present invention will be further described in detail below with reference to the embodiments. The following description is only for explaining the present invention and does not limit its content.

[0034] The technical solutions used to achieve the purpose of the present invention are:

[0035] The preparation and application of the detection probe for liver cancer marker GP73 of the present invention. The preparation method of the probe includes the following steps:

[0036] 1. Induced expression of target protein GP73

[0037] The addition of IPTG induces protein expression because it binds to the repressor protein and changes the protein conformation, which leads to inactivation of the repressor protein and transcription. Too short induction time will result in less expression of target protein; too long induction time will result in transcriptional expression of non-target genes. Therefore, it is necessary to explore an optimal induction time to obtai...

Abstract

The invention belongs to the technical field of genetic engineering, immunology and oncology, particularly relates to preparation and application of a probe for detecting a liver cancer marker GP73, and discloses GP73 antibody modified nanogold. An enzyme linked immunosorbent assay based on a nanogold probe is adopted to be used for a signal amplification strategy of high sensitivity detection ofan antibody against GP73. The probe is formed by high load capacity of an HRP enzyme-marked goat anti human IgG antibody on the surface of the nanogold, the stability is good, and preparation is easy.In the process of detection, firstly a soluble hepatocellular carcinoma associated antigen GP73 is purified by the expression of genetically engineering bacteria, and a protein is used as an antigento correspondingly detect hepatocellular carcinoma, hepatitis, liver cirrhosis, a benign tumor of liver and the level of the antibody against GP73 in normal human serum. Wider application prospects intumor diagnosis, tumor prognosis and detection evaluation are achieved, compared with a traditional ELISA method, the positive serum concentration detected by the probe is lower, and the sensitivityof the method is higher.

Description

Technical field [0001] The invention belongs to the technical fields of genetic engineering, immunology and oncology, and particularly relates to the preparation and application of a detection probe for the liver cancer marker GP73. Background technique [0002] Liver cancer is one of the most common malignant tumors in clinic, ranking fifth in malignant tumors. Liver cancer is also a high incidence in my country. There are about 110,000 new liver cancer cases every year, accounting for about 40% of the world's cases. It has become a major killer that seriously threatens the health and lives of our people. A large number of studies and experiments have proved that the most effective way to control the incidence of liver cancer, prevent the occurrence of cancer, and reduce the mortality rate is to achieve early detection, early diagnosis and early treatment. However, because the early symptoms and signs of liver cancer are not obvious or specific, an ideal early diagnosis method ...
