Abstract

The invention discloses cryoprotectant capable of being used for hematopoietic stem cells or lymphocytes and a cryopreservation method. The cryoprotectant is prepared from dimethyl sulfoxide (DMSO), human serum albumin and normal saline in proportioning and blending modes, and the volume ratio of the DMSO to the human serum albumin to the normal saline is 1 to 2 to 2. The invention further comprises a method for cryopreservation of the hematopoietic stem cells or the lymphocytes by applying the cryoprotectant, and the method comprises the following steps of (1) cryoprotectant preparation, (2)cryopreservation subpackage, (3) heat sealing and (4) gradient cryopreservation. A cell culture medium and animal serum are not added into the cryoprotectant, thus infusion can be directly conducted,and no adverse reaction application is achieved; compared with traditional cryoprotectant, allergen bringing is avoided, and safety is higher; the cryoprotectant is high in cryopreservation efficiency, the activity of the stem cells after cryopreservation resuscitation is greater than 85%, the activity of the lymphocytes is greater than 90%, and the manufacturing and using costs of the cryoprotectant are lowered on the basis that the physiological properties of the cryopreserved cells are effectively maintained. The cryopreservation method is easy and convenient to operate, required equipmentis simple, and the cryopreservation method is suitable for being applied in different levels of hospitals.
