Embodiment 1

[0097] A method for preparing targeted anti-tumor T cells, comprising the following steps:

[0098](1) Prepare the gene sequence of the chimeric antigen receptor CAR-IL13RA2 targeting IL13RA2

[0099] The genes encoding the signal peptide, the single-chain antibody targeting IL13RA2, the hinge region of CD8α, the transmembrane region of CD8, the 4-1BB signal region and the CD3ζ signal region were respectively prepared. As shown in NO: 22, the coding gene of the single-chain antibody targeting IL13RA2 is shown in SEQ ID NO: 3, the coding gene of the CD8α hinge region is shown in SEQ ID NO: 10, and the coding gene of the CD8 transmembrane region is shown in As shown in SEQ ID NO: 12, the coding gene of the 4-1BB signal region is shown in SEQ ID NO: 14, and the coding gene of the CD3ζ signal region is shown in SEQ ID NO: 16.

[0100] The genes encoding the above-mentioned signal peptide, single-chain antibody targeting IL13RA2, CD8α hinge region, CD8 transmembrane region, 4-1BB ...

Abstract

The invention provides a targeting antitumor T cell. The targeting antitumor T cell comprises an IL13Ralpha2-targeting chimeric antigen receptor CAR-IL13Ralpha2 and/or an HER2-targeting chimeric antigen receptor CAR-HER2, wherein the CAR-IL13Ralpha2 comprises amino acid sequences of an IL13Ralpha2-targeting single chain antibody, an extracellular hinge region, a transmembrane region and an intracellular signal region, which are sequentially connected from an amino terminal to a carboxyl terminal, and the CAR-HER2 comprises amino acid sequences of an HER2-targeting single chain antibody, an extracellular hinge region, a transmembrane region and an intracellular signal region, which are sequentially connected from the amino terminal to the carboxyl terminal. The targeting antitumor T cell can efficiently recognize and kill tumor cells of surface IL13Ralpha2 and/or HER2 positive antigens, can effectively overcome immune escape of tumor cells, and has lasting tumor killing effects. The invention also provides a preparation method and an application of the targeting antitumor T cell.

Description

technical field [0001] The invention relates to the field of medical biology, in particular to a targeted anti-tumor T cell and its preparation method and application. Background technique [0002] Glioma, short for glioma, is the most common malignant tumor of the central nervous system (CNS), originating from glial cells. Glioma is a tumor with strong invasiveness and high recurrence rate. Due to its invasive growth, surgical treatment is difficult to remove all tumor cells. Even if it is supplemented with radiotherapy and chemotherapy, recurrence is still inevitable. [0003] Chimeric Antigen Receptor T-Cell Immunotherapy (CAR-T) technology developed in recent years is one of the more effective treatments for malignant tumors. CAR-T technology injects a new solution for adoptive cellular immunotherapy by utilizing the targeting, killing activity and persistence of T cells expressing tumor-specific chimeric antigen receptors through genetic modification technology.
