Embodiment 1

[0033] Embodiment one, material and method

[0034] 1. The microbial samples to be screened come from the Shandong Provincial Marine Microbial Resource Center.

[0035] 2. Medium

[0036] TSB medium (g / L): tryptone 17g, soybean peptone 3g, sodium chloride 5g, dipotassium hydrogen phosphate 2.5g, glucose 2.5g.

[0037] Grease neutral red medium (g / L): 6ml of olive oil, 20ml of Tween-80, 5g of sodium chloride, 2g of potassium dihydrogen phosphate, 0.1g of anhydrous magnesium sulfate, 1.5g of dipotassium hydrogen phosphate, 1g of ammonium sulfate , Potassium nitrate 1g, 0.8% neutral red aqueous solution 2ml, agar 18g, pure water 1000ml, pH7.2, sterilized at 121°C for 20min.

[0038] Fermentation medium (g / L): olive oil 6ml, Tween-80 20ml, sodium chloride 5g, potassium dihydrogen phosphate 2g, anhydrous magnesium sulfate 0.1g, dipotassium hydrogen phosphate 1.5g, ammonium sulfate 1g, potassium nitrate 1g, 1000ml pure water, pH7.2, sterilized at 121℃ for 20min.

Embodiment 2

[0051]Example 2, Screening and identification of Pseudomonas monteilii (Pseudomonas monteilii) XZL-13

[0052] A strain of microorganism that can use oil to grow was screened through the oil neutral red medium, which was identified as Pseudomonas monteilii, and the strain was named XZL-13. Inoculate the purely cultured XZL-13 strain in TSB medium and culture it at 25°C. The colony is milky yellow and raised, with irregular edges and smooth surface ( figure 1 ). Gram-stained strains are red and are Gram-negative bacteria. Electron micrographs show that it is a short rod with flagella ( figure 2 ). The results of physiological and biochemical tests showed that the strain could utilize sodium citrate and produce naphthol-AS-BI-phosphatase and urease (Table 1).

[0053] Pseudomonas monteilii (Pseudomonas monteilii) XZL-13 was deposited in General Microorganism Center of China Committee for Culture Collection of Microorganisms, CGMCC; the deposit number is CGMCC No.17514.

Abstract

The application has screened and obtained a microorganism that can utilize oil. The microorganism is Pseudomonas monteilii (Pseudomonas monteilii) XZL-13, which is preserved in the General Microorganism Center of China Microbiological Culture Collection Management Committee, and the preservation number is CGMCC No. 17514. The optimum temperature of the microorganisms is 28°C, the optimum pH value is 7.0, the optimum salinity is 2g / L, and the time to reach the maximum culture biomass is 7h. It has the corresponding ability to use 7 kinds of edible oils including corn oil, animal oil, peanut oil, sesame oil, coconut oil and olive oil.

Description

technical field [0001] The application relates to the technical field of microorganisms, in particular to Pseudomonas monsonii for degrading oil and its application. Background technique [0002] With the improvement of people's living standards and the development of the tertiary industry in cities, my country's catering industry is increasing at a rate of more than 10% per year. As an important catering raw material, edible oils and fats are used more and more. The degree of application of waste edible oils and the harm caused by returning to the food market have attracted people's attention. [0003] Statistics show that my country's annual output of waste oil is as high as 5 million tons. Taking Beijing and Tianjin as an example, it reached about 300,000 tons in 2016. Moreover, once the waste edible oil enters the environment as a pollutant, it will cause immeasurable environmental pressure. [0004] At present, the ways to deal with waste edible oils include anaerobic...
