Abstract

The invention discloses a method for determining diethylstilbestrol by adopting up-conversion luminescence immunochromatography. The method is characterized in that according to the research, a solvothermal method is adopted to synthesize up-conversion nanoparticles (UCNPs)-NaYF4: Yb, Er, surface functionalization modification is carried out, the nanoparticles are coupled with a diethylstilbestrol(DES) monoclonal antibody to prepare a fluorescent probe, a nitrocellulose film is respectively sprayed on a bovine serum albumin-diethylstilbestrol (BSA-DES) conjugate and a goat anti-mouse secondary antibody to form a test strip detection line (T line) and a quality control line (C line), and an up-conversion luminescence immunochromatographic test strip is established for rapid quantitative detection; a method of DES is adopted; the experimental result shows that the DES linear quantitative detection range of the test strip is 25 to 10000 ng/mL (Y is equal to 0.43927 x-0.57647, and R2 is equal to 0.996), the detection limit is 20.84 ng/mL, the detection time of a single sample is 15 min, intra-batch and inter-batch variable coefficients are both smaller than 10%, the specificity recognition capacity is high, an average value of RSD detection values is about 15% after the kit is stored at 37 DEG C for 7 days; standard recovery experiments show that the average recovery rate is 90.1%-115.2%, relative standard deviation is smaller than 5%, and the method has good consistency with high performance liquid chromatography.
