Problems solved by technology

The disadvantages of this model are as follows: 1) The normal and undegenerated intervertebral disc is used to compress the nerve root, which is inconsistent with the clinical reality of lumbar disc herniation, and the pain symptoms caused by it are relatively mild and the duration is relatively short (only about 1 -2 weeks); 2) There are few pain behavioral evaluation methods based on rats; 3) There are few transgenic rat strains available for research, which is not conducive to in-depth research on the biological mechanism of pain

Abstract

The invention discloses a construction method of a lumbar intervertebral disc protrusion mouse model. The method comprises the following steps of 1) performing lumbar intervertebral disc puncture through an abdominal approach to induce lumbar intervertebral disc degeneration; 2) feeding animals for 4 weeks to obtain degenerated lumbar intervertebral disc nucleus pulposus tissues; 3) exposing spinal canal dural sacs and nerve roots through a posterior approach, and placing degenerated intervertebral disc tissues in an intervertebral foramen area to form nerve root compression, so as to obtain the lumbar intervertebral disc protrusion mouse model; and 4) performing behavioral detection to evaluate the performance and degree of lower limb radiation pain. The degenerated intervertebral disc tissues are used for pressing the nerve roots, severe and lasting lower limb radiation pain symptoms can be induced, and the chronic disease course of the clinical lumbar intervertebral disc protrusionis well simulated; and the mouse is used as a model animal, so that a plurality of objective and credible pain behavioral measurement methods and rich transgenic strains are beneficial to deep exploration of a neurobiological mechanism of the lower limb radiation pain of the lumbar intervertebral disc protrusion.

Description

technical field [0001] The invention relates to the field of biomedicine, in particular to a method for constructing a mouse model of lumbar disc herniation. Background technique [0002] Lumbar disc herniation is one of the main causes of lumbago and lower extremity radiating pain, which is quite common in clinic. Lumbar and leg pain symptoms caused by lumbar disc herniation seriously impair the quality of life and work ability of patients, and affect economic production. At present, the neurobiological mechanism of pain caused by lumbar disc herniation has not been fully elucidated. An effective and credible animal model of lumbar disc herniation is a key scientific research tool to further elucidate the neural mechanism of low back pain and develop new clinical drugs. [0003] Early related studies mostly used larger animals such as pigs, dogs or rabbits to make models of lumbar disc herniation. These large animal models are expensive and relatively convenient to opera...
