Problems solved by technology

[0006] In other current techniques, the actual sperm separation is not possible due to the time constraints of the sample (such as obtained by testicular surgery), but only sperm purification
The sperm purification process is usually more time-consuming; sorting sperm from a testicular sample is a manual operation, typically takes 2-3 hours, and only removes the seminal plasma from the sperm (World Health Organization, 2010)
[0007] This means that this sperm preparation method, and other current sperm preparation methods, may not be able to pick the most fertile sperm from the sample

Embodiment Construction

[0081] The following provides detailed descriptions of the embodiments of the present invention to help those skilled in the art implement various aspects of the present invention.

[0082] Various embodiments of the devices, methods of isolating sperm, methods of screening sperm, and methods of fertilization described herein can be practiced in a fertility clinic or research center to study in vitro fertilization (IVF), intrauterine insemination (IUI) and oocyte Application of intracytoplasmic sperm injection (ICSI) in the treatment of male infertility. The information disclosed herein can be used to isolate motile sperm and subsequently screen the sperm for IVF, ICSI, IUI, and / or other reproductive treatments. The devices and methods, and variants and methods thereof, may also be used in non-human applications, particularly agriculture, such as assisted reproductive technology (ART) treatment in cattle and other animal breeding.

Abstract

The invention provides a device and method for separating active sperms from a seminal fluid sample. The device comprises an inlet reservoir for receiving the seminal fluid sample, an outlet reservoir for collecting sperms separated from the sample, and at least one, but preferably a plurality of, micro-channels disposed between the inlet reservoir and the outlet reservoir to provide fluid communication between the two reservoirs. The micro-channel comprises at least one wall defining a boundary surface along which sperm is directed to the outlet reservoir, and the at least one wall has at least one surface structure selected from, for example, one or more recesses, protrusions, and combinations of the recesses and protrusions. The surface structure extending over at least a portion of the length of the microchannel to provide at least one additional boundary surface along which the active sperm travels towards the outlet reservoir.

Description

technical field [0001] The present invention relates to a device and method for separating spermatozoa from a semen sample, and more particularly to a device and method for selecting spermatozoa with predetermined characteristics from the separated spermatozoa. Background technique [0002] Sperm selection is an important component of assisted reproductive technology (ART), with a strong impact on the success of reproductive treatments and the health of embryos (Davies, 2012 and Mcdowell, 2014). Much of the focus of reproductive therapy and biology has revolved around egg retrieval and embryo culture, yet the importance of the health of male gametes and their isolation strategies has been grossly underestimated since the successful in vitro fertilization (IVF) in 1973. [0003] Recent studies (Oseguera-López, 2019 and Sakkas, 2015) have described the important and complex role of sperm beyond the transmission of the second set of chromosomes. However, current clinical sperm...
