Abstract

The method is based on high-throughput sequencing and comprises the following steps of (1) extracting whole genome DNA (Deoxyribonucleic Acid) in chicken blood, constructing a gene library, and sequencing by utilizing a second-generation sequencing technology; (2) screening the high-quality sequencing read segments by referring to a mitochondrial whole genome sequence of a related species, removing nuclear DNA fragments, splicing the mitochondrial genome, selecting an optimal splicing result, and predicting and annotating encoding protein genes, rRNA and tRNA of the spliced mitochondrial genome; and (3) carrying out PCR (Polymerase Chain Reaction) amplification on the target fragment to finally establish a library, and then carrying out double-terminal sequencing on the library by using Illumina NovaSeq.
