The invention comprises application of the dried indigofera stachyoides root extract in preparation of the whitening and freckle-removing preparation, application of the dried indigofera stachyoides root extract as an inhibitor to inhibit tyrosinase activity and proliferation of melanoma cells, application of the dried indigofera stachyoides root extract to inhibit synthesis of melanin, and a method for separating and purifying the dried indigofera stachyoides root extract from dried indigofera stachyoides root. According to the method for separating and purifying the dried indigofera stachyoides root extract, the purity of total flavonoids of dried indigofera stachyoides root can be improved to 46% or above. The dried indigofera stachyoides root extract provided by the invention can be used as the inhibitor for inhibiting the activity of tyrosinase, inhibiting the proliferation of melanoma cells, inhibiting the activity of tyrosinase in the melanoma cells and synthesizing melanin, and has very good whitening and freckle-removing effects.
