Embodiment

[0019] In this example, in order to solve the problem of difficult clinical wound healing, a preparation method of leukocyte-platelet-rich plasma (leukocyte-platelet-rich plasma, L-PRP) was explored with great concentration. The platelets in the L-PRP prepared in this example The content is about 4 times that of whole blood, and it is rich in leukocytes and IL-6. It has been found through experiments that L-PRP can activate STAT3 by up-regulating the transcription level of IL-6, promote the migration of keratinocytes, and accelerate wound re-epithelialization. For specific methods, see below.

[0020] See attached figure 1 ,in figure 1 A and figure 1 B is a schematic diagram of the preparation process of the PRP of the present embodiment, and the specific preparation process is as follows:

Abstract

The invention relates to the technical field of blood products, and discloses a preparation method of leukocyte-rich and platelet-rich plasma (L-PRP), comprising the steps of sampling blood and preparing L-PRP; the invention also discloses an application of the L-PRP in wound treatment. The platelet content of the prepared L-PRP is about 4 times that of whole blood, the L-PRP is rich in leukocytes and interleukin 6 (IL-6), and it is found that the L-PRP can promote keratinocyte migration and accelerate wound reepithelization by up-regulating the IL-6 transcriptional level and activating signal transduction and transcriptional activation protein 3 (STAT3); according to the prepared L-PRP and the research on the L-PRP, classification and application scenes of platelet-rich plasma (PRP) are refined, a new mechanism of promoting wound healing by the L-PRP is found, a new solution is provided for clinically solving refractory wounds, and the L-PRP has important clinical practical significance.

Description

technical field [0001] The invention relates to the technical field of blood products, in particular to a preparation method of leukocyte-rich platelet-rich plasma. Background technique [0002] Refractory wounds refer to the phenomenon of delayed or interrupted physiological healing due to insufficient angiogenesis, denervation, cell migration disorders, etc., and are generally defined as wounds with a healing time of more than 3 months. Many diseases can affect wound healing. With aging, smoking, obesity, diabetes and other problems all over the world becoming more serious, the incidence of difficult-to-heal wounds is gradually increasing. This will not only cause pain, infection, and even disability, amputation, and psychological problems for patients, but also bring huge economic burdens to society. Therefore, how to promote wound healing has become an important proposition for clinicians to solve urgently. [0003] Wound epithelialization Restoration of an intact skin...
