Problems solved by technology

Traditional bacterial or yeast systems sometimes fail to produce complex proteins in functional form
Although some mammalian cells can replicate complex proteins, they are often difficult or expensive to culture and produce only relatively low amounts of protein
In addition, non-secreted proteins are relatively difficult to purify from prokaryotic or mammalian cells since they are often not secreted into the culture medium

[0148] transgenic mammal

[0149] Detailed methods for making non-human transgenic animals are described here and in the "Examples" section below.

[0150] The method may include introducing the DNA construct into the germline of the mammal to produce a transgenic mammal. For example, one or a few copies of the construct can be integrated into the genome of a mammalian embryo by standard transgenic techniques.

[0151] While cattle and goats are preferred, other non-human mammals can also be used. Preferred non-human mammals are ruminants such as cows, sheep, camels or goats. Other examples of preferred non-human mammals include horses, pigs, rabbits, mice and rats. For nuclear transfer techniques, the mammal used as a source of cells, eg, genetically engineered cells, will vary depending on the transgenic mammal desired to be obtained. For example, the bovine genome should be used for nuclear transfer of bovine oocytes.

Abstract

The invention provides transgenically produced decorin and methods of making and using transgenically produced decorin.

Description

[0001] This application claims the benefit of provisional application 60 / 201,932, filed May 5, 2000, the contents of which are hereby incorporated in their entirety. Background of the invention [0002] An increasing number of recombinant proteins are being developed for therapeutic, diagnostic, agricultural, veterinary, nutritional and other uses; however, many of these proteins can be difficult or expensive to produce in functional form in large quantities using conventional methods. [0003] Conventional methods often involve inserting the genes responsible for the production of specific proteins into host cells such as bacteria, yeast or mammalian cells. These cells are grown in culture medium and the protein of interest is recovered from the cells or culture medium. Traditional bacterial or yeast systems sometimes fail to produce complex proteins in functional form. Although certain mammalian cells can replicate complex proteins, they are often difficult or expensive to ...
