Said invention uses the high-pressure pump to make the phospholipid dissolved in organic phase, liposoluble medicine and aqueous solution be quickly mixed in solution mixer to obtain multiphase multilayer liposome, then makes said multiphase multilayer liposome directly pass through liposome extruding device so as to obtain large monochamber liposome, and the medicine which is not wrapped in the liposome and organic solvent can be removed by using high-pressure ultrafiltering dialysis column.
