The method for improving the success rate of umbilical cord blood mesenchymal stem cell culture by using placental mesenchymal stem cells provided by the invention shortens the culture time of the placental mesenchymal stem cells, effectively solves the problem that the placental mesenchymal stem cells are low in purity and poor in multiplication capacity and the like, improves the success rate of umbilical cord blood mesenchymal stem cell culture and develops the utilization value of the umbilical cord blood mesenchymal stem cells. As fetal calf serums and pancreatin are not used, the safety of the cells on clinical application is enhanced.

The invention provides a separation and culture method of renal podocyte of a mouse. The separation and culture method comprises the following steps of (a), intraperitoneally injecting 2% pentobarbital sodium to execute the mouse, sterilizing the mouse with alcohol, taking out kidney tissue of the mouse, putting the kidney tissue into a precooled sterile PBS (Phosphate Buffer solution) containing double antibodies, soaking and washing the kidney tissue, and peeling and removing a renal capsule; (b), mechanically dissociating the kidney tissue, then digesting the kidney tissue by a preheated mixed enzyme solution, and terminating digestion by a mixed complete culture solution; (c), obtaining the renal podocyte by adopting a difference sieving method, inoculating the podocyte in a treated cell bottle, and culturing the podocyte; (d), digesting a cell, which is cultured for 3d, through pancreatin, then filtering the cell once again by a sieve mesh, centrifuging filtrate to remove supernatant fluid, and resuspending and inoculating an obtained mixture in the treated cell culture flask; (e), identifying the renal podocyte by cell immunofluorescence. The separation and culture method of the renal podocyte of the mouse, which is provided by the invention, is good in repeatability, and is simple and convenient to operate; moreover, the high-yield and high-purity renal podocyte of the mouse can be obtained.

In comparison research with crustose coralline algae, the survival rate and the adhesion rate of the coral larvae treated by the method are higher than those of the coral larvae treated by the crustose coralline algae under the same treatment. And after 24 hours of treatment with calcium chloride, the growth ability of the subsequent coral larvae is not affected. Meanwhile, under the condition that the survival and adhesion rate of the coral larvae are guaranteed, calcium ions are used for inducing the coral larvae to adhere efficiently and stably, and calcium chloride is more convenient and quicker to add in an experiment, is simple to operate and is better controlled as a single variable. Therefore, the method also has the potential of large-scale engineering application.
