Abstract

This invention relates to molecular cloning of uncoupling protein (UCP) of Lampetra japonica. The UCP gene is cloned from Lampetra japonica with degenerate primer through 3',5'-RACE. UCP gene is associated with serveral diseases, such as diabetes mellitus, cardiovascular diseases and tumors. UCP gene can be used as a drug target for manufacturing drugs for treating above diseases.

1. Technical field [0001] The invention belongs to the field of biotechnology, and in particular relates to molecular cloning of lamprey uncoupling protein (uncoupling protein, UCP). 2. Background technology [0002] The main function of mitochondria is to participate in energy metabolism. When mitochondria respire, the respiratory chain transfers electrons and pumps protons from the mitochondrial matrix to the membrane space, forming the mitochondrial transmembrane potential. Protons return to the mitochondrial matrix by driving ATP synthase to synthesize ATP, forming a coupling of oxidation and phosphorylation. UCP localized in the mitochondrial inner membrane can leak protons from the mitochondrial intermembrane space back into the matrix, thereby reducing the membrane potential of mitochondria and uncoupling oxidation and phosphorylation.
