The method is divided into the following three steps: 1, condensing brain olefine aldehyde and butanone by adopting potassium hydroxide as a catalyst and aqueous solution of methanol as solvent to prepare 3-methyl-5-(2,2,3-trimethyl-3-cyclopentene-1-base)-3-pentene-2-ketone; 2, subjecting the 3-methyl-5-(2,2,3-trimethyl-3-cyclopentene-1-base)-3-pentene-2-ketone and methyl iodide to addition reaction by adopting octadecyl trimethyl ammonium bromide as the catalyst in order to prepare 3,3-dimethyl-5-(2,2,3-trimethyl-3-cyclopentene-1-base)-4-pentene-2-ketone; and 3, reducing the 3,3-dimethyl-5-(2,2,3-trimethyl-3-cyclopentene-1-base)-4-pentene-2-ketone by adopting potassium borohydride as a reducing agent so as to prepare the polysantalol.
