Problems solved by technology

[0004] At present, most of the detection methods of Aeromonas hydrophila and Aeromonas temperatus are traditional physiological and biochemical identification and immunological diagnosis. Because many of their biochemical characteristics are unstable and the serotypes are also complicated, when using the above methods to identify prone to misjudgment
There are also methods for rapid detection of bacterial species-specific genes using PCR technology, but only one of them can be detected
Since a total pathogenic bacteria content is required when judging the possibility of pathogenicity, two different kits and two PCR tests are required for the same sample to obtain the total data of the two bacteria. In terms of epidemic forecast, prevention, control, etc., it is not fast enough

Embodiment Construction

[0018] 1. A kit for detecting Aeromonas hydrophila and Aeromonas temperatus by FQ-PCR, consisting of a packaging box and packaged reagents, each reagent is hermetically packaged in a reagent bottle. The packaged reagent list is as follows:

[0019] One 4 μL upstream primer of 4 μM, the DNA sequence of the upstream primer is 5’-TGG CCT TGA CAT GTC TGG AAT CCT-3’;

[0020] 4μL of 4μM downstream primer, the DNA sequence of the downstream primer is 5'-ACC ATT ACG TGC TGG CAA CAA AGG-3';

[0021] 4 μL of 2 μM TaqMan probe, the DNA sequence of the TaqMan probe is 5’-(FAM)-AAT CAG AAC ACA GGT GCT GCA TGG CT-(TAMRA)-3’.

[0022] 2. In addition to the reagents in the above example, there are also packaged in the kit of this example

[0023] One 100μL bottle of 2X Premix EX Taq reaction solution;

[0024] One bottle of sterile double distilled water 68μL.

[0025] 3. In addition to the above two reagents, the kit in this case also contains

[0026] One 0.1mL rapid DNA extraction re...

Abstract

The invention provides a kit for detecting aeromonas hydrophila and aeromonas sobria through fluorescence quantitive polymerase chain reaction (FQ-PCR), and comprises an upstream primer of which the DNA sequence is 5'-TGGCCTTGACATGTCTGGAATCCT-3', a downstream primer of which the DNA sequence is 5'-ACCATTACGTGCTGGCAACAAAGG-3', and a TaqMan probe of which the DNA sequence is 5'-(FAM)-AATCAGAACACAGGTGCTGCATGGCT-(TAMRA)-3', wherein each reagent is packaged in a sealed mode. The kit is designed according to 16SrDNA in the aeromonas hydrophila and aeromonas sobria; and compared with the prior art, the kit has the advantages that: only one pair of universal primers is adopted to simultaneously detect the two pathogenic bacteria, and the real-time and quick effect is achieved in the aspects of emergency pre-warning and burst disease treatment.

Description

technical field [0001] The invention relates to a kit for detecting Aeromonas hydrophila and Aeromonas temperis by FQ-PCR, which belongs to the technical field of determination or inspection of enzymes or microorganisms in chemistry. Background technique [0002] Aeromonas hydrophila ( Aeromonas hydrophila ) is widely distributed in nature, and generally exists in fresh water, sewage, silt, soil and human feces, and can infect fish, amphibians, reptiles, birds and mammals, and humans can also be infected by this fungus. Food poisoning, diarrhea, affects food safety, and is an opportunistic pathogen for immunosuppressed patients and patients with liver function diseases. It is a typical pathogenic bacteria of humans, animals and fish. In addition to causing huge economic losses to the aquaculture industry , and its public health significance, especially in terms of clinical diseases and food safety, has been increasingly valued by relevant departments. [0003] Aeromonas so...
