Embodiment Construction

[0061] The present invention discloses a new method and device for using stem cells. These stem cells can be derived from cord blood or peripheral blood (but not mesenchymal). These cells can be isolated and expanded using simple techniques. A particularly useful aspect of this invention is that the cells may be isolated from an individual, particularly an adult individual, for autologous stem cell therapy, or the cells may be isolated from another individual for non-autologous stem cell therapy. The present invention also discloses the use of stem cells in regulating the function of mononuclear cells, such as T lymphocytes, B lymphocytes, monocytes, dendritic cells (DC) and granulocytes.

[0062]In a preferred embodiment, these stem cells have the following characteristics, including but not limited to the following characteristics, expressing stem cell markers Oct-4, Nanog and Sox-2, and other genes related to embryonic stem cells, such as zinc finger (Zinc finger) and SCA...

Abstract

Methods and apparatus are disclosed co-culturing stem cells with mononuclear cells and / or lymphocytes to modulate their function. The invention also discloses the use of stem cells to educate autoreactive immune cells as a mechanism to treat autoimmune diseases and immune disorder-related diseases, such as diabetes. In one aspect of the invention, bioreactors are disclosed for modulating lymphocytes and suppressing autoreactive T cells. The bioreactors can include a chamber having at least one positively charged and / or hydrophobic substrate surface, a population of stem cells attached to the substrate surface, an inlet conduit for introducing lymphocytes into the chamber, and an outlet conduit for extracting treated lymphocytes following co-culturing with the stem cells.

Description

[0001] Cross references related to this application [0002] This patent application claims the benefit of U.S. Provisional Patent Application No. 61 / 283,782, filed December 8, 2009, entitled "Stem Cell Educators and Clinical Applications"; and U.S. Provisional Patent Application No. 61 / 283,810, filed On December 8, 2009, titled "Stem Cell Immunomodulation and Its Molecular Mechanisms," these findings were integrated into the current patent application to form a single entity. This patent application, to a certain extent, is also a continuation of another US patent application, application number 12 / 099,054, filed April 7, 2008, titled "Isolation of human umbilical cord blood-derived embryonic-like stem cells", and the same discovery Integrate with the current patent application to form a single entity. technical field [0003] The present invention relates to general methods and devices for treating autoimmune diseases and diseases related to immune disorders.
