Embodiment Construction

[0012] The methods of molecular biology used in the following examples of the present invention are all known techniques.

[0013] The protein encoded by the Oyster oyster HMGB gene in the present invention, the amino acid sequence of the protein is shown in SEQ ID NO:2. The nucleotide sequence of the coding frame of the gene encoding the aforementioned protein is shown in SEQ ID NO.1.

[0014] The application of the protein in the preparation of the oyster anti-infection immune preparation of the present invention is to immunize New Zealand white rabbits with the protein to obtain polyclonal antiserum. The polyclonal antiserum (i.e. polyclonal rabbit antibody to HMGB protein) is used to inhibit the inflammatory response, cell necrosis and apoptosis caused by the pathogen RLO of Oyster oyster and the cell wall component of Gram-negative bacteria - lipopolysaccharide (LPS). Death.

[0015] Implementation steps of the present invention include:

[0016] 1. Using the total RNA...

Abstract

The invention relates to the field of biology and aims to provide application of high mobility group box (HMGB) protein and antibody to the preparation of an ostrea rivularis anti-infection immune preparation. The amino acid sequence of the protein is shown as the SEQ ID NO:2; the coding frame nucleotide sequence of the gene is shown as the SEQ ID NO:1; the application of the protein to the preparation of the ostrea rivularis anti-infection immune preparation is to immunize New Zealand white rabbits by using the protein so as to obtain polyclonal antiserum. According to the invention, the ostrea rivularis HMGB gene coding frame full sequence is obtained; the soluble HMGB protein product is expressed and purified by establishing a prokaryotic expression vector; and the polyclonal rabbit antiserum is prepared. The polyclonal rabbit antiserum has the effect of inhibiting inflammatory response caused by ostrea rivularis pathogeny RLO and gram negative bacteria LPS as well as necrobiosis and apoptosis. The invention relates to the field of biology and aims to provide application of high mobility group box (HMGB) protein and antibody to the preparation of an ostrea rivularis anti-infection immune preparation. Description

technical field [0001] The invention belongs to the field of biotechnology, and relates to HMGB gene, protein and its function in preparing oyster anti-infection immune preparation. Background technique [0002] High mobility group box B (high mobility group box, HMGB for short) is a member of the high mobility group box protein family. Structurally, HMGB contains three characteristic domains, namely two HMG boxes (A box and B box) with similar structure and high homology, and an acidic C-terminal rich in aspartic acid and glutamic acid. HMGB acts as a "DNA chaperone" in the nucleus, and can participate in life activities such as DNA recombination, repair, gene transcription regulation, cell replication, differentiation and maturation by interacting with various transcription factors, replication proteins and steroid receptors. In addition, the current research shows that HMGB can be released outside the cell under specific conditions and mediate multiple inflammatory respo...
