Problems solved by technology

At the same time, different batches of silkworms of the same species with the same genetic characteristics have different developmental processes in the process of seed production and temperature-sensing treatment, and it takes a lot of labor to dissect all the different batches using conventional detection methods, so the establishment of corresponding labor-saving Methods to monitor progress of embryonic development are needed

Embodiment 1

[0028] Example 1: A method for measuring the developmental progress of silkworm embryos

[0029] ⑴Materials and equipment:

[0030] The silkworm eggs were bred by Suzhou University (Suxiu×Chunfeng), the greening control box was a 250D artificial climate box produced by Guangdong Shaoguan Xinteng Science and Technology Instrument Co., Ltd., and the balance was one ten-thousandth gram produced by Shimadzu Corporation of Japan. Precision electronic balance, quantitative PCR instrument is ABI7300.

[0031] Sample: Take 50mg eggs from each area, set up 3 replicates, and set up 5000 parallel areas for anatomical identification of developmental stages.

[0032] ⑵Activation, temperature sensing and greening

[0033] Activation is naturally released at low temperature, and the sensitive temperature is 15°C for 10 days, and the greening is processed according to the simplified greening technology.

Abstract

The invention discloses a method for testing the development process of silkworm embryos. The method specifically comprises the following steps of: (1), establishing a model expressing the incidence relation between the relative transcription expression quantity of the acetylcholine esterase type 1 (ace1) of a to-be-tested variety of the silkworm embryos and the development process of the embryos; (2), selecting at least one embryo from the to-be-tested silkworm embryos to use as a sample, testing the relative transcription expression quantity of the ace1 of the sample, and then contrasting with the model obtained in step (1) to determine the development process of the silkworm embryos according to the relative transcription expression quantity of the ace1 of the to-be-tested sample. The development stage of a whole graine can be accurately determined by the method through the test on the development process of a small quantity of silkworm embryos. Manual operation errors can be reduced, the test process is simplified, and a development process identification method is provided for molecular biology study of the silkworm embryos.

Description

technical field [0001] The invention relates to a method for measuring the developmental progress of silkworm embryos, in particular to an accurate detection method for the developmental progress of silkworm embryos based on the relative transcriptional expression of genes. Background technique [0002] The silkworm (Bombyx mori L.) has been domesticated in my country for more than 5,700 years. It belongs to the complete metamorphosis insect of the family Lepidoptera, and its generations include egg, larva, pupa and adult (moth). It takes about 13-14 days for silkworm eggs to hatch from activation to hatching. In order to achieve the goal of silkworm eggs hatching neatly, ant silkworms strong and timely hatching, within 13-14 days, the environmental conditions should be adjusted according to the progress of embryonic development. This operation process is referred to as quickening. [0003] In order to facilitate the identification of different development stages of silkwor...
