The invention discloses a method for detecting acetylcholin esterase by using a multi-hot-spot active targeting type SERS (Surface Enhanced Raman Scattering) substrate, which comprises the following steps: soaking the multi-hot-spot active targeting type SERS substrate in a solution to be detected, taking out after sufficient time, and carrying out SERS detection; the preparation method of the multi-hotspot active targeting type SERS substrate comprises the following steps: preparing a gold nanosphere superlattice two-dimensional membrane, synthesizing gold and silver double-component hollow nanoparticles, attaching the gold nanosphere superlattice two-dimensional membrane to a silicon wafer, modifying a beta-cyclodextrin-methylene blue inclusion compound, and adding the gold and silver double-component hollow nanoparticles. According to the method, the beta-cyclodextrin-methylene blue inclusion compound-gold nanosphere superlattice two-dimensional film-silicon wafer can be prepared firstly, after the beta-cyclodextrin-methylene blue inclusion compound-gold nanosphere superlattice two-dimensional film-silicon wafer is combined with an object to be detected, the gold-silver double-component hollow nanoparticles are added, and SERS detection is conducted. When the method is used for detecting the acetylcholin esterase, the sensitivity, the accuracy and the repeatability of a detection result can be improved.

Preparation method and application of novel acetylcholin esterase activated curcumin-based fluorescent probe for detecting diazinon

The invention discloses a preparation method and application of a novel acetylcholin esterase activated curcumin-based fluorescent probe for detecting diazinon. The fluorescent probe is 4, 6-bis [(1E)-2-(4-(((dimethylamino) carbonyl) oxy)-2-methoxyphenyl) vinyl]-2, 2-difluoro-2H-1, 3, 2-dioxo boron heterocyclic hexene tetrafluoroborate, which is called YPC for short. The probe YPC can specifically recognize acetylcholin esterase, after acetylcholin esterase is added, orange fluorescence of a solution is remarkably enhanced, meanwhile, due to the inhibition effect of diazinon on the activity of acetylcholin esterase, the orange fluorescence of the probe is quenched, and therefore detection of diazinon is achieved. The detection limit of the probe YPC to acetylcholin esterase is as low as 5.2114 mU / mL, the detection limit of the probe YPC to diazinon is as low as 1.0463 ng / mL, and the probe YPC shows a good linear relationship in the range of 0-12 ng / mL.

The invention discloses an application of a scopolamine model in an acetylcholin esterase inhibitor, and relates to the technical field of medicines.The application comprises the steps that S1, the learning memory of the scopolamine model is improved through a compound; s2, the inhibition effect of the compound on acetylcholin esterase is verified, and the compound is specifically (5S)-1-(3, 4-dihydroxyphenyl)-5-hydroxy-7-(4-hydroxy-3-methoxyphenyl) heptyl-3-ketone. A plurality of groups of experiments are designed to prove that the compound can remarkably improve the learning and memory dysfunction of AD model mice induced by scopolamine and has dose dependence, an enzyme activity inhibition experiment shows that the compound has strong inhibitory activity on AChE, the half inhibitory concentration (IC50) of the compound reaches 5.5 nM and is more remarkable than that of a clinical first-line anti-AD drug donepezil (IC50 = 12.2 nM), and the compound can be applied to preparation of anti-AD drugs. As the AChE activity increase is a key factor of cholinergic system injury in the AD pathological process, the compound is speculated to reduce hydrolysis of acetylcholine by inhibiting the AChE activity, and further improve the learning and memory ability of model animals.
