The application discloses a preparation method and application of gold nanometer bipyramid@ZIF-8 wrapped curcumin co-loaded nano-preparation, and comprises the following steps: S1, gold nanometer bipyramid is prepared and photoresponsive ligand modification is carried out; S2, the gold nanometer bipyramid is dispersed in a reaction system containing zinc salt, polyvalent metal salt, organic imidazole ligand, curcumin and defect inducer to form a mixed system; S3, double-wavelength near-infrared light irradiation is applied to induce in-situ growth of ZIF-8 layer doped with polyvalent metal ions on the surface, and a multi-level defect structure is formed; S4, a second active substance is loaded; S5, a biological enzyme responsive linker or an active targeting ligand is modified; and S6, a biological source polymer material is coated. Through synergistic regulation of photoinduced in-situ growth and defect engineering, the application realizes controllable growth and multi-level defect construction of the ZIF-8 layer, and the prepared nano-preparation is uniform in structure, closely combined in interface, stable in double active substance loading, excellent in enzyme-like catalytic activity, and can be used for preparation of tumor treatment drugs.
