The preparation method comprises the following steps: firstly, preparing silver colloid as a raw material for standby; secondly, carefully cleaning a glass slide; thirdly, self-assembling a single layer of polydipropylene dimethyl ammonium chloride with positive charge on the surface of the glass slide; fourthly, adding sliver nano granules of a pre-aggregation raw material with moderate NaCl into the silver colloid; and finally soaking the glass slide assembled with the positive charge into the pre-aggregation silver colloid solution, and self-assembling and forming a single layer of compact silver granules on the surface of the glass slide to obtain the single-layer silver nanoparticle film with the high surface enhanced Raman scattering activity.
