Disclosed is a cervico-vaginal tissue equivalent comprised of vaginal epithelial cells and immune cells, cultured at the air-liquid interface. The tissue equivalent is capable of being infected with a sexually transmitted pathogen such as a virus (e.g., HIV), a bacteria, a helminthic parasite, or a fungus. The tissue equivalent is also capable of undergoing an allergic-type reaction or an irritant-type reaction. The tissue equivalent is characterized as having nucleated basal layer cells and nucleated suprabasal layer cells, and further as having cell layers external to the suprabasal layer progressively increasing in glycogen content and progressively decreasing in nuclei content. Immune cells of the tissue equivalent are primarily located in the basal and suprabasal layers. Also disclosed are methods for producing the tissue equivalent. The methods involve providing vaginal epithelial cells and immune cells, seeding the cells onto a porous support, and co culturing the seeded cells at the air-liquid interface under conditions appropriate for differentiation. One such method disclosed is for generation of the tissue equivalent in serum free medium. Specific cells from which the tissue equivalent is generated, and also specific preferred components of the medium in which the tissue equivalent is generated are provided. Also disclosed is a cervico-vaginal tissue equivalent produced by the methods disclosed herein.

Improved topsheet contacting hydrous body tissues, and absorbent device using the topsheet

The present invention relates to an improved body contacting surface or topsheet for an absorbent device which is in contact with the aqueous tissue of the user's body. The aqueous body tissue may include, but is not limited to, aqueous tissue such as interlabial skin or tissue and mucosal tissue such as vaginal tissue. In one non-limiting embodiment, the topsheet comprises a moderately hydrophilic to substantially hydrophobic nonwoven web that is not absorbent. The topsheet is disposed on absorbent devices such as sanitary napkins, tampons, panty liners, interlabial devices, incontinence devices, bandages, and other articles. The present invention also relates to a method of retaining exudates from a source of exudates in a user. wherein at least a portion of the exudate source is located within an aqueous film having an irregular surface, such as the interlabial or intravaginal region of a female user. The present invention also relates to an absorbent article insertable into the interlabial region of a female user, the absorbent article comprising an absorbent storage core and a body-contacting vaginal discharge-receiving layer disposed over a on the storage core. Needle-punched communication is provided between the absorbent storage core and the body-contacting exudates-receiving layer. Additionally, the absorbent storage core includes a unitary web of fibrous material and may optionally include an absorbent gelling material and a plurality of perforations. The absorbent article also includes an insertion device, which may be an absorbent article inserter or a foldable reinforcing tongue integral with the absorbent article, the reinforcing tongue having fold lines.
