The present invention relates to a selectively soluble polymer capable of binding to a desired biomolecules in a mixture containing various biological materials and the methods of using such a polymer to purify a biomolecule from such a mixture. The polymer is soluble in the mixture under a certain set of process conditions such as pH or temperature and / or salt concentration and is rendered insoluble and precipitates out of solution upon a change in the process conditions. The polymer is capable of binding to the desired biomolecule (protein, polypeptide, etc) and remains capable of binding to that biomolecule even after the polymer is precipitated out of solution. The precipitate can then be filtered out from the remainder of the stream and the desired biomolecule is recovered such as by elution and further processed.

Polymerization process and materials for biomedical applications

A molded component or article for biomedical use is prepared from a crosslinkable non-water-soluble polymer which when crosslinked and saturated with water forms a hydrogel. The polymer is formulated as a composition containing a non-aqueous diluent in addition to the polymer, the diluent being present in a volumetric proportion that is substantially equal to the volumetric proportion of water in the hydrogel that would be formed when the polymer is crosslinked and saturated with water. The composition is cast in a mold where the composition is exposed to conditions that cause crosslinking to occur by a reaction to which the non-aqueous diluent is inert. The crosslinking reaction produces a molded non-aqueous gel which is then converted to a hydrogel by substituting an aqueous liquid such as water or physiological saline for the non-aqueous diluent. The use of a molding composition whose curing consists essentially entirely of crosslinking results in a molding process that entails little or no shrinkage, and dimensional integrity is maintained up through the formation of the hydrogel by using the non-aqueous diluent in essentially the same volumetric proportion as water in the hydrogel.

C-, S- and N-glycosylation of peptides

The present invention provides polypeptide conjugates wherein a modifying group such as a water-soluble polymer, a therapeutic agent or a biomolecule is covalently linked to the polypeptide through a glycosyl linking group. In one embodiment, the polypeptide includes a glycosylation consensus sequence, wherein glycosylation occurs at an aromatic amino acid residue, such as the C-2 or the N-1 position of a tryptophan side chain. Process for the fractionation of polymers

A process for the purification or fractionation of aqueous soluble polymers using an aqueous two-phase system is described. The concentrations of the polymer to be fractionated and of an aqueous soluble salt, and the temperature of the aqueous fractionation medium are adjusted so that two phases form, the lower molecular weight polymer molecules partition into the high salt concentration phase, and the higher molecular weight polymer molecules partition into the low salt concentration phase. The resulting high molecular weight polymers are characterized by a higher average molecular weight and a narrower molecular weight distribution and decreased unsaturation than the unfractionated polymers. After being subjected to the fractionation process, polyol polymers that form hydrogels in aqueous solution exhibited higher viscosities and a liquid to gel transition over a narrower temperature range than the unfractionated polyol polymers.
