The invention relates to an application of autologous adipose-derived mesenchymal stem cells in preparation of medicines for treating joint diseases. The application is characterized in that adipose-derived mesenchymal stem cells (Ad-MSCs) separated and extracted from autologous adipose tissues of the patient are injected to the diseased articular cavity in a large dose once. The adopted mesenchymal stem cells have immunophenotyping and representing biological marks of Ad-MSC through flow cytometry authentication, and can be directionally induced and differentiated to osteoblast and chondrocyte. Primary cells are directly injected to the articular cavity of the diseased joint to treat various joint diseases. With the adoption of the autologous adipose-derived mesenchymal stem cells as the primary cells, the cells have better differentiative potential and the ability of inhibiting inflammation and immunoregulation ability. The adipose-derived mesenchymal stem cell transplant provides a novel effective means for treating osteoarticular diseases. The method is simple and feasible, and the quantity of cells collected is great and the trauma is small. The autologous mesenchymal stem cells of the patient are applied without invading related ethics, so that the risk of immunological rejection, infectious diseases and the like is avoided.

Mesenchymal stem cell preparation for repairing articular cartilage injury or coloboma, preparation method and application thereof

The invention discloses a mesenchymal stem cell preparation for repairing articular cartilage injury or coloboma, a preparation method and application thereof. According to the preparation method, mesenchymal stem cells and synovial cells are indirectly cocultured, so as to obtain the mesenchymal stem cells after coculture, and then the mesenchymal stem cells are cryopreserved, umbilical cord mesenchymal stem cells, which are acquired after the synovial cells and the umbilical cord mesenchymal stem cells are inoculated at density ratio of 4:1 and cocultured, are easier to differentiate towardcartilage direction under a same induction condition. The preparation method utilizes a constructed animal knee joint cartilage injury or coloboma model to prove that the umbilical cord mesenchymal stem cells cocultured with the synovial cells have an obvious effect of facilitating the repairing of articular cartilage injury or coloboma. The invention further provides a preparation which is prepared from the cocultured mesenchymal stem cells. The preparation can be applied to clinic repairing of articular cartilage injury or coloboma; the preparation can effectively promote the repairing of articular cartilage injury or coloboma through anticular injection; the therapeutic effect of the preparation is obviously better than that of a pure mesenchymal stem cell preparation.

High-strength gradient full-absorption artificial ligament and preparation method thereof

The invention discloses a high-strength gradient full-absorption artificial ligament and a preparation method of the high-strength gradient full-absorption artificial ligament. The artificial ligamentcomprises an articular cavity free section and marrow channel woven sections at the two ends of the articular cavity free section, wherein the marrow channel woven sections are of sheath-core double-layer tube-shaped structures, and the sheath-core double-layer tube-shaped structures are composed of sheath layers and core layers. The preparation method comprises the following step: preparing themarrow channel woven sections of the sheath-core double-layer tube-shaped structures by adopting a weaving, knitting or braiding integrated forming technology, wherein weft yarns are not arranged at the articular cavity free section, namely, parallel free yarns not participating in weaving are adopted. The adopted yarns are formed by twisting and compounding fibers with different degrading periodsat the specific ratio, after implantation, a three-dimensional spiral through porous channel can be constructed through gradient degrading, thus the oriented growth of host cells is induced, and meanwhile, the requirement for the mechanical property needed during the tendon-bone healing process is met.
