Embodiment 1

[0037] Embodiment 1. extracts the method for total alkaloids from the leaking reed flower

[0038] 1.1 Preparation of experimental materials

[0039] The leaking reed flower, collected from Daqing Mountain, Hohhot City, Inner Mongolia, was identified as the dried flower of the genus Rhaponticum uniflorum (L.) DC.) in the subfamily Asteraceae Tubular Flower Subfamily Qizhou.

[0040] 1.2 Specific methods of the experiment

[0041] 1.2.1 Preparation of buffer solution and chromogenic reagent

[0042] pH=5.4 buffer solution: Take 40mL of 0.1mol / L citric acid solution and 85mL of 0.1mol / L sodium citrate solution in a 250mL volumetric flask, and dilute to the mark with distilled water to obtain the solution.

[0043] Bromocresol Green Indicator Solution: Weigh 0.125g of bromocresol green, dissolve in 25mL 0.1mol / L NaOH solution, add 2.55g of potassium hydrogen phthalate, dissolve in water, transfer to a 250mL volumetric flask, and dilute to volume with water Instantly.

Embodiment 2

[0054] Embodiment 2. single factor test

[0055] Four factors including ultrasonic temperature, ultrasonic power, solid-liquid ratio and ultrasonic time were selected for single factor experiment, and the relationship between each factor and the extraction rate of total alkaloids was obtained.

[0056] 2.1 The effect of ultrasonic temperature on the extraction rate of total alkaloids from Leu reed flower

[0057] Precisely weigh 0.5 g of Leuhua, and perform ultrasonic extraction at 20, 30, 40, 50, and 60 °C with ultrasonic power of 200 W, solid-liquid ratio of 1:30 g / mL, and ultrasonic time of 30 min. After being treated according to the color development method in Example 1, the absorbance was measured. The results are shown in Table 2.

Embodiment 3

[0072] Embodiment 3 single factor test result analysis

[0073] 3.1 The effect of different ultrasonic temperatures on the extraction rate of total alkaloids in Leu reed flower, the results are shown in image 3 , when the ultrasonic temperature is 30°C, the extraction rate drops sharply. After 30°C, the image takes the shape of a parabola. At the ultrasonic temperature of 30, 40, and 50°C, the extraction rate of alkaloids is higher, and the extraction rate is the highest at 40°C. Therefore, 30, 40, and 50°C were selected for response surface analysis.

[0074] 3.2 The effect of different ultrasonic power on the extraction rate of total alkaloids in Leu reed flower, the results are shown in Figure 4 . The whole image shows a monotonous increase, but the increasing trend is slow. This is because the greater the ultrasonic power, the stronger the ultrasonic effect, the more total alkaloid leached matter, and the greater the extraction rate. At 160, 180, and 200W, the extract...

Abstract

The invention researches ultrasonic extracting technology of total alkaloids in rhaponticum uniflorum. Content measurement is performed. The amount of the alkaloids in the rhaponticum uniflorum is measured through ultraviolet spectrophotometry and serves as evaluation indexes, and therefore an optimal process can be determined. According to the optimal extracting technology, achieved after response faces are optimized, of the total alkaloids in the rhaponticum uniflorum, ultrasonic temperature is 41 DEG C, ultrasonic power is 200 W, the ratio of materials to liquid is 1:28 g / ml, ultrasonic time is 40 min, and the four-factor influence sequence is the ultrasonic power, the ratio of the materials to the liquid, the ultrasonic temperature and ultrasonic time in sequence, the content of the total alkaloids is 0.2774%, the average recovery rate is 99.77%, and the RSD is 2.0% (n=9). The alkaloid extracting technology with the responses faces optimized is high in precision and predictability and provides a scientific and reasonable theoretical basis for further development.

Description

technical field [0001] The invention relates to the technical field of bioengineering, in particular to an ultrasonic extraction method for extracting total alkaloids from succulents. Background technique [0002] Rhaponticum uniflorum (L.) DC., a common Chinese Mongolian medicine, is also known as the dried flower of Rhaponticum uniflorum (L.) DC. It belongs to the genus Rhaponticum uniflorum (L.) DC. As a commonly used Mongolian medicine, Leuluhua has the effects of relieving tingling, clearing away heat, detoxifying, and relieving the exterior. [0003] The total alkaloids are the main medicinal components of Leuhua. For this reason, it is of great significance to study the extraction, separation and purification methods of the total alkaloids in Leuca japonica for the follow-up research on their biological activities. [0004] In order to fully obtain the total alkaloids from S. spp., the cell structure of S. spp. should be destroyed as much as possible. It is an inno...
