Embodiment Construction

[0034] In order to make the technical problems, technical solutions and advantages to be solved by the present invention clearer, the following will describe in detail with reference to the drawings and specific embodiments.

[0035] An application of CPT-II in the process of malignant transformation induced by fat accumulation hepatocytes, comprising the following steps:

[0036] (1) Grouping of animals: male Sprague-Dawley (SD) rats of clean grade, 4-6 weeks old, and weighing 100-120 g, randomly selected 10 as the control group, and then 6 male Sprague-Dawley rats as the control group One group was randomly divided into 8 fatty liver groups and 8 cancer induction groups;

[0037] (2) Model preparation: the rats in the fatty liver group were fed with high-fat diet, and the rats in the cancer-inducing group were fed with high-fat diet plus 0.05% 2-fluorenyl acetamide (2-FAA, Sigma, USA) Feed with granule feed, take 1 normal mouse, a group of fatty liver mice and a group of tu...

Abstract

The invention provides application of CPT-II in the malignant transformation process induced by fat accumulation of liver cells. The application comprises the following steps of animal grouping, model preparing, blood enzymology and lipid detecting, liver cell fat staining, liver tissue protein preparing and concentration determining, quantitative analysis on the liver CPT-II concentration, pathologic histology examining, liver CPT-II immuohistochemistry staining and statistical analyzing. According to the application, by establishing fatty liver and liver mouse models and detecting changes of the CPT-II in the liver cell malignant transformation process, it is found that the CPT-II content in the fatty liver model is obviously lower than that of a normal control group, it shows that decrease of the liver cell CPT-II can be caused by liver lipid accumulation and fat degeneration, the CPT-II content of a liver cancer mouse model is significantly lower than that of the fatty liver model, and therefore the CPT-II content is in negative correlation with the liver cell malignancy grade. The liver CPT-II specific concentration and immunohistochemistry prove that the liver cells are progressively decreased in the canceration process, it shows that fat accumulation of the liver cells is aggravated due to low expression or dysfunction of the CPT-II, and a promoting effect is achieved on the liver cell maglignancy transformation process.

Description

technical field [0001] The invention belongs to the field of medical research, development and utilization, and specifically relates to the application of CPT-II in the process of malignant transformation induced by fat accumulation liver cells. Background technique [0002] Nonalcoholic fatty liver disease (NAFLD) is a clinical syndrome characterized by diffuse macrovesicular fatty degeneration of liver cells and excessive liver lipid accumulation caused by alcohol and other definite liver damage factors. Pathological syndromes, including simple fatty liver and evolving steatohepatitis and cirrhosis, are divided into mild, moderate and severe fatty liver according to the degree of liver fat exceeding liver weight. NAFLD has become the leading cause of chronic liver disease in Europe, the United States, Japan and other countries, and it is also one of the main risk factors for the occurrence of hepatocellular carcinoma (HCC) in addition to hepatitis B virus (HBV) and hepatit...
