Abstract

A method for improving the vigor of endosperm of Nepal pauliata seeds after low-temperature storage, comprising: taking out the dry seeds stored at low temperature from -18°C to -20°C together with the airtight container for storing the seeds; The seeds were placed in an incubator at a temperature of 45°C for 24 hours, and the seeds were taken out after heat treatment for 6 hours. After the seeds returned to room temperature, they were placed on 1% agar medium, and placed in a 20°C culture room for 24 hours; the treated seeds were cut through the seed coat and endosperm, soaked in 1% triphenyltetrazolium chloride (TTC ) solution, placed in the dark at 30°C for 48 hours; the soaked seeds were taken out and cut to observe the endosperm staining, and the endosperm with vitality showed a uniform red color. The method significantly improves the endosperm vigor of the Nepal pauliata seeds stored at low temperature for different periods of time in different places of production. Solved the problem that the vigor of endosperm decreased after cryopreservation of wild pauliata seeds in Nepal.
