Embodiment 1

[0020] 1. Materials and methods:

[0021] 1.1 Research materials:

[0022] In July 2013, mature fresh Mallotus barbatus seeds were collected in Changlong Village, Nanping Township, Shangsi County, Fangchenggang City, Guangxi. After collection, they were transported back to the Southwest Wild Organism Germplasm Bank Seed Preservation Center of Kunming Institute of Botany.

[0023] 1.2 Research methods:

[0024] 1.2.1 Drying and refrigerating and returning to temperature after refrigerating: put the seeds collected in step 1.1 in a drying room at 15°C and 15% relative humidity to dry. Regularly use the Hygrolab C1unit relative humidity measuring instrument produced by Rotronic Ltd. combined with the Rotronic HC 2-AW probe produced by the same company to measure the relative humidity of the seeds, and put them into a closed container after the equilibrium relative humidity reaches 15%. On November 11, 2014, the airtight container containing the seeds was placed in a refrigerat...

Embodiment 2

[0036] 1. Materials and methods:

[0037] 1.1 Research materials:

[0038] On August 13, 2012, mature fresh Mallotus barbatus seeds were collected in Jiaoshi Town, Fuling District, Chongqing. After collection, they were transported back to the Southwest Wild Organism Germplasm Bank Seed Preservation Center of Kunming Institute of Botany.

[0039] 1.2 Research methods:

[0040] 1.2.1 Drying and refrigerating and returning to temperature after refrigerating: put the seeds collected in 1.1 in a drying room at 15°C and 15% relative humidity to dry. On August 9, 2013, the airtight container with the seeds was placed in -18°C to -20°C for refrigeration. Remove the container with...

Abstract

The invention discloses a method for improving the activity of mallotus barbatus seed embryos preserved at a low temperature. The method comprises the following steps: taking out dried seeds preserved at a low temperature ranging from minus 18 DEG C to minus 20 DEG C together with a closed container for storing the seeds; putting the dried seeds and the closed container in an environment with the temperature of 15DEG C and the relative humidity of 15 percent for 24 hours, and taking out the seeds; putting the seeds into an incubator with the temperature of 45DEG C, carrying out heat treatment for 6 hours and then taking out; restoring the temperature of the seeds to room temperature, putting the seeds on a 1 percent agar culture medium, and putting the seeds in a culturing room with the temperature of 20DEG C for 24 hours; cutting seed coats and endosperms of the treated seeds, soaking the treated seeds into 1 percent triphenyltetrazolium chloride (TTC) solution, and putting a mixture in a dark condition with the temperature of 30DEG C for 48 hours; taking out the soaked seeds, cutting off and observing dyeing conditions of the embryos, and displaying uniform red color for the embryos with the activity. According to the method disclosed by the invention, the problem that the activity of the embryos is reduced after mallotus barbatus seeds are preserved at low temperature is solved; in addition, the treatment method is simple and feasible and is free of risk and toxic and / or side effects.

Description

technical field [0001] The invention belongs to the technical field of plant germplasm resource preservation, and in particular relates to a method for improving the vigor of paulownia seed embryos preserved at low temperature (-18°C to -20°C). Background technique [0002] Mallotus barbatus (Wall.) Muell. Arg. is a small tree species of Euphorbiaceae (Euphorbiaceae) Mallotus Lour.. Distributed in Eastern and Southern Asian countries. It is mainly produced in Yunnan, Sichuan, Guizhou, Hunan, Guangdong and Guangxi in China. Born in forest margins or shrubs at an altitude of 400-1 300 meters. [0003] The stem bark fiber can be used as raw material for papermaking; the wood is light and soft and can be used for making utensils; the seed oil can be used as industrial oil. At present, there is no research report on the germination and storage of tung tree seeds. [0004] Drying normal seeds and storing them at low temperature (-18°C to -20°C) can greatly prolong their lifesp...
