Abstract

A method for improving activity of seed endosperm of mallotus apelta after low temperature preservation comprises: taking out dried seeds in low temperature preservation at -18 DEG C to -20 DEG C and a sealed container storing the seeds; placing same in an environment with the temperature of 15 DEG C, and relative humidity of 15% for 24 hours, and taking out the seeds; placing the seeds in an incubator at the temperature of 45 DEG C, performing heat treatment for 4-6 hours, and then taking out the seeds; after recovering to the room temperature, placing the seeds on an agar culture-medium of 1%, and placing same at a culture chamber at 20 DEG C for 24 hours; cutting the peel and endosperm of the processed seeds, soaking in a 1% triphenyltetrazolium chloride (TTC) solution, and placing at a dark condition at 30 DEG C for 48 hours; taking out the soaked seeds, cutting same to observe dyeing conditions of the endosperm, wherein the endosperm having activity shows even red. The method obviously improves the activity of endosperm of the mallotus apelta seeds from different producing areas and stored at a low temperature for different times, and solves the problem that the endosperm of the mallotus apelta seeds is reduced after low temperature preservation. Moreover, the processing method is simple and easy to achieve without any danger or toxic and side effects.
