Abstract

The invention discloses a method for improving the vitality of endosperms of mallotus nepslensis seeds preserved at low temperature. The method comprises the following steps: taking out dry seeds preserved at the low temperature of 18DEG C below zero and 20DEG C below zero together with a closed container for storing the seeds; putting the seeds in an environment with the temperature of 15DEG C and relative humidity of 15 percent for 24 hours, and taking out the seeds; putting the seeds in an incubator with the temperature of 45DEG C, carrying out heat treatment for 6 hours and then taking out the seeds; restoring the temperature of the seeds to room temperature, then putting the seeds on an agar culture medium with the concentration of 1 percent, and putting the agar culture medium in a culturing room with the temperature of 20DEG C for 24 hours; cutting off seed coats and the endosperms of treated seeds, soaking the cut seeds in a 1 percent triphenyltetrazolium chloride (TTC) solution, and putting the mixture under the dark condition of 30DEG C for 48 hours; taking out soaked seeds, cutting the seeds to observe the endosperm dyeing condition, wherein the endosperms with the vitality show uniform red colors. According to the method, the vitality of the endosperms of the mallotus nepslensis seeds which are from different production places and are preserved at low temperature is remarkably improved; the problem that the vitality of the endosperms of the mallotus nepslensis seeds stored at the low temperature is reduced is solved.
