Abstract

The invention discloses a method of simultaneously detecting residues of different types of veterinary drugs in fish meat. In the method, a sample is subjected to pretreatment with a mixture solutioncomposed of acetonitrile, methanol and water according to the volume ratio of 3:1:1 and contains 1% of acetic acid (v/v) and 10 mM of EDTA disodium. A purification operation is composed of one time oflow temperature standing treatment and two times of low-temperature high-speed centrifugation, so that majority of co-extracted protein and lipid impurities are removed. Finally, qualitative detection and quantitative analysis to all target substances are completed by means of tandem of ultra-high performance liquid chromatography and Orbitrap high-resolution mass spectrum under positive and negative ionization modes. The method can simultaneously detecting eighty kinds, covering twelve types, of common fish-use veterinary drug compounds, thus achieving general detection of the veterinary drug compounds in fish meat at high sensitivity, broad spectrum, high speed and high throughput. The method includes simple and high-effective pre-treatment process, so that extraction efficiency of theresidual compounds in various types is ensured. Tandem ultra-high performance liquid chromatography and Orbitrap high-resolution mass spectrum represents high selectivity and sensitivity, so that screening and quantification to potential target substances in the sample can be completed at a lower level.
