[0087] Molecules that specifically bind other molecules are essential for many biomedical and analytical applications, such as therapeutics, diagnostics, laboratory research and many aspects of analytical science. The present disclosure provides many significant advantages. The present disclosure allows repeated rounds of selection using populations of candidate molecules of considerable length. The present disclosure relates to novel methods of DNA display for the discovery and evolution of binding ligands for biomedical and analytical applications. These binding partners include, but are not limited to, nucleic acids or proteins (including, but not limited to, antibodies, antibody fragments, peptides, polypeptides). In addition to selecting binding ligands, DNA display is also used to select and improve catalytic activity, including but not limited to enzymes, ribozymes, and aptamymes.

Abstract

Described herein is a method that displays RNA on the DNA from which it was encoded, enabling enhanced selection of RNA aptamers and proteins. The covalent link between RNA and the DNA which encodes it can be used to negate the spatial informational loss following transcription. This method has applications in the selection of binding ligands such as RNA aptamers and antibodies as well as catalytic molecules such as aptazymes and enzymes.

[0001] 1 Introduction [0002] The present disclosure relates generally to DNA display, methods of making DNA display libraries, methods of using DNA display or DNA display libraries to select ligands that bind to a target. Described herein are methods for preparing DNA display libraries for displaying RNA transcribed from DNA templates. The method involves the process of linking nucleoside triphosphates (RNA) to oligonucleotides (DNA) via a bridging linker molecule. Also described herein are methods for capturing transcripts (RNA) and ligating them to their encoding templates (DNA). Also described herein are methods of attaching transcripts (RNA) via their encoding template (DNA) to a scaffold such as a solid support or beads.
