Abstract

The invention discloses a recombinase polymerase isothermal amplification and test strip combined method for detection of Staphylococcus aureus. The method includes the steps of: designing a specificStaphylococcus aureus primer pair, taking the Staphylococcus aureus genomic DNA as the template, conducting recombinase polymerase isothermal amplification, and applying a colloidal gold test strip toacquire the detection result. The method is rapid and simple, has no need for expensive instruments and equipment, can generate a detectable level product only through 10min amplification under a 40DEG C constant temperature condition, and does not need a tedious electrophoresis process, a result can be acquired within 5min of colloidal gold test strip detection, and the result can be observed bynaked eyes. In addition, the method has strong specificity, is free of reaction with other bacterial genuses, has high sensitivity, and makes up for the disadvantages of the existing traditional detection technology. The method is expected to be applied to grass-roots inspection and quarantine institutions and become a conventional detection means, and is of important significance for improving food hygiene level, ensuring food safety and promoting the international food trade development.
