[0086] 1. Experimental materials

[0087] 1. Main reagents

[0088] Sodium bisulfite and hydroquinone were purchased from Sigma, and Trizol was purchased from Invitrogen. Wizard DNA purification kit was purchased from Promega. PCR amplification kits were purchased from Sangon Bioengineering (Shanghai) Co., Ltd.

[0090] The specimens were all from the biological specimen bank of Wuxi City People's Hospital between May 2015 and May 2017. A total of 45 patients with renal cancer were operated, ranging in age from 52 to 70 years old, with a median age of 63 years. All patients were not treated with chemotherapy and radiotherapy before surgery. All subjects in this study signed the informed consent form and were approved by the hospital ethics committee. The primary tumor tissue and the adjacent normal tissue more than 5cm away...

Abstract

The invention discloses a gene detection kit for detecting a chromophobe cell carcinoma subtype in renal carcinoma. The gene detection kit comprises a primer for detecting complete methylation at 74bp-165bp sites of a gene ENST00000571911. The 74bp-165bp sites of the gene ENST00000571911 are completely methylated in a primary lesion of the chromophobe cell carcinoma, while paracancerous normal tissue of the chromophobe cell carcinoma and primary lesions and paracancerous normal tissue of the other three subtypes are all unmethylated, so that the chromophobe cell carcinoma subtype can be diagnosed by detecting whether the 74bp-165bp sites of the gene ENST00000571911 are completely methylated; the specificity is strong; the diagnostic accuracy rate of 45 samples in the experiment is 100%.

Description

technical field [0001] The invention belongs to the field of gene diagnosis and relates to a gene detection kit for detecting subtypes of chromophobe cell carcinoma in renal cancer. Background technique [0002] In clinical practice, the incidence of renal cell carcinoma (remahcell carcinoma, RCC) has been increasing year by year, and has become a high-incidence and common malignant tumor of renal parenchyma. Previous studies have shown that the disease mostly occurs in people aged 50 to 70, and only about 5% of patients are under 40 years old. However, in recent years, the number of young RCC patients has increased year by year. RCC tumors contain multiple subtypes, and there are differences in pathological manifestations and prognosis between each subtype. Accurate pathological typing of them at an early stage can effectively improve the prognosis of patients, improve the effect of anticancer treatment and the survival rate of patients ( Reference: Comparative analysis of...
