Embodiment 1

[0024] Embodiment 1: A kind of seedling stage identification method of nitrogen-efficient varieties of rice

[0025] 1) Select a healthy and plump variety to be tested, soak the seeds at 25-35°C for 2 days, and accelerate germination at 40°C for 1 and a half days. After the germination, the seeds are sown in washed sand without adding any nutrients and cultivated to 2 leaves and 1 heart;

[0026] 2) Take seedlings with uniform growth, wash them with water and transfer them to nitrogen-free culture medium (the formula is as follows: 0.32 mmolNaH 2 PO 4 • 2H 2 O, 1.0 mmol K 2 SO 4 , 1.0mmol CaCl 2 , 1.7mmol MgSO 4 •7H 2 O, 9.1×10 -3 mmol MnCl 2 • 4H 2 O, 5.2×10 -4 mmol (NH 4 ) 6 MoO 24 • 4H 2 O; 1.8×10 -2 mmol H 3 BO 3 , 1.5×10 -4 mmol ZnSO 4 •7H 2 O, 1.6×10 -4 mmol CuSO 4 • 5H 2 O. 3.6×10 -2 mmol FeCl 3 •6H 2 O), after 1 week of culture, transfer to the control medium (formulation as follows: 0.29 mmol NH 4 NO 3 , 0.32 mmol NaH 2 PO 4 • 2H 2 O, 1....

Abstract

The invention relates to a method for identifying rice nitrogen efficient varieties in a seedling stage. The method comprises the following steps: selecting a healthy and full variety to be tested, soaking seeds for 2 days at the temperature of 25-35 DEG C, accelerating germination for one day and a half at the temperature of 40 DEG C, and then seeding in cleaned sand without any nutrients and culturing until seedlings have 2 leaves and 1 heart; taking the seedlings which uniformly grow, washing with clear water and then transferring into a nitrogen-free medium; after one week of culture, transferring the seedlings into a control medium and an N-feedback medium for culturing; after that, measuring the areas LA of the rice leaves as well as the corresponding dry matter weight LDW, root weight RW and root number RN; determining a nitrogen physiological feedback value so as to determine whether the rice is the nitrogen efficient variety or not, wherein the higher the nitrogen feedback index is in the seedling stage, the higher the nitrogen utilization rate of the rice is. By establishing a specific hydroponic system and determining the nitrogen feedback index of the seedling stage bymeasuring the leaf and root indexes of the rice in the nursery stage, the method can quickly and efficiently evaluate the nitrogen efficient utilization characteristic of the rice.

Description

technical field [0001] The invention relates to a seedling stage identification method for rice nitrogen-efficient varieties. Background technique [0002] Low nitrogen use efficiency has always been a prominent problem in my country's agricultural production, although a large number of studies have shown that there are a large number of available genetic resources with high nitrogen utilization in the rice germplasm resource bank. However, due to the close relationship between the absorption and utilization of nitrogen and the formation of yield, and it is easily affected by environmental effects; on the other hand, the evaluation and screening indicators of nitrogen efficiency often need to observe the entire growth period to obtain the final results. And varieties, there are defects such as long cycle and heavy workload, and there are a lot of uncertain factors, such as climate, soil environment, diseases and so on. Therefore, the simplified methods for the identificatio...
