Problems solved by technology

But species identification by morphological methods has always been limited
First, phenotypic plasticity affects morphological traits and leads to errors in species identification
Second, because some key morphological features are only applicable to specific life stages, many individuals cannot be accurately identified
However, in the prior art, there are no reports on the SNP molecular markers and applications thereof for identifying the short-jawed anchovy, lake anchovy, and sword anchovy according to the present invention

[0076] A total of 20 Anchovy samples were collected from 2 lakes, Poyang Lake (PY, n=10, from three sampling points) and Dongting Lake (DT, n=10, from three sampling points). Twenty lake anchovy samples were collected from two lakes, Taihu Lake (TH, n=10, from three sampling points) and Chaohu Lake (CH, n=10, from two sampling points). The 12 migratory Anchovy samples were from the main channel and important tributaries of the Yangtze River (SH, n=12), such as Jingjiang, the Yangtze River tributary around Chongming Island, and Luchaogang along the coast of Shanghai (Table 1).

[0077] Samples from multiple collection points were selected from each lake to measure the Sr / Ca ratio of trace elements in sagittal otoliths (4-5 samples per lake), and an...

Abstract

The invention relates to the field of biotechnology, and specifically discloses SNP molecular markers for identifying short-jawed anchovy, lake anchovy and sword anchovy and applications thereof. The inventor obtained 120 SNP sites with significant differences between the short-jawed anchovy and the sword anchovy and 21 difference sites between the lake anchovy and the sword anchovy in the previous screening. The present invention selects 20 sites with a differentiation index of 1 among the 120 sites for primer design; and selects 10 designed primers for sites with a higher differentiation index among the 21 sites. The results of the study showed that: Anchovy brevignathus and Anchovy chinensis can be completely separated by using any one of the above 19 SNP molecular markers. However, 8 of the 10 SNP molecular markers used to distinguish the anchovy from the anchovy can be amplified in the tested samples and used for species identification. The invention provides a stable, high-efficiency and low-cost method for identifying the short-jawed anchovy, the anchovy, the anchovy and the anchovy, and provides an effective tool for the species identification and resource protection of the anchovy in the future.

Description

technical field [0001] The invention relates to the field of biotechnology, in particular to SNP molecular markers for identifying short-jawed anchovy, lake anchovy and sword anchovy and application thereof. Background technique [0002] In my country, the anchovy is mainly divided into two ecological types, namely the river-sea migratory type and the lake-dwelling type. However, the classification of sedentary shortjaw anchovy, lake anchovy and migratory anchovy has been controversial. Anchovy is a fish with high economic value. Driven by huge profits, many businessmen use short-jawed anchovy and lake anchovy to pass off as anchovy from the Yangtze River. How to distinguish between short-jawed anchovy, lake anchovy and sword anchovy has always been a hot spot of research by many scholars. [0003] Early taxonomic research provided a morphological basis for the identification of different ecotypes of Anchovy. It is convenient, simple and intuitive to identify species based...
