Embodiment 1

[0021] The technical approach provided by the present invention to solve the above-mentioned technical problems:

[0022] ①Phronoid screening, cleaning and ecological element recording of the population gene bank

[0023] Records collected in Yushan Islands, Ningbo, Zhejiang Province: Great White Reef was selected, with latitude and longitude, facing waves, mid-tide, and facing northeast, on December 22, 2014, January 20, 2015, February 17, 2015, respectively. Laver was collected separately on March 19, 2015. The environmental elements and ecological conditions (temperature, salinity, pH, etc.) of the field investigation were recorded respectively, as shown in Table 1 below.

Abstract

The invention provides a porphyra haitanensis group thallus germplasm bank construction method to increase diversity of germplasm resources, simplify operation methods and reduce the management cost. The method comprises the steps as follows: airing cleaned thalli, and performing quick drying and dehydration until the dehydration rate is 80%-90%; placing the thalli in sealing bags with 20-30 thalli as a preservation unit, pumping out air in the sealing bag for sealing, and preserving the thalli in a dark place at subzero 20-subzero 24 DEG C for 3-5 years. By means of the porphyra haitanensis group thallus germplasm bank construction method based on living body preservation, in the following preservation procedure, living bodies can be preserved under the condition of sealing and shading atthe low temperature of subzero 20-subzero 24 DEG C within 3-5 years without other operation.

Description

technical field [0001] The invention belongs to the technical field of algae species preservation, and in particular relates to a method for constructing a thallus germplasm bank of algae porphyra population based on living body preservation. Background technique [0002] Laver is the cultivated seaweed with the highest economic value in the world today, and the cultivation of laver also has good social and ecological benefits. Altar laver is not only distributed naturally in the coasts of Zhejiang and Fujian in my country, but also the most important cultivated economic algae in this sea area. With the expansion of cultivation scale, self-crossing and inbreeding in artificial seedlings lead to premature maturity and germplasm degradation in cultivated offspring. At the same time, wild resources in the natural environment have also experienced population degradation, biomass reduction, and natural provenance simplification. Therefore, it is urgent to preserve germplasm res...
