Abstract

The invention discloses a method for detecting correlation between a gene GARNL1 and cockscomb and wattle traits of a cock and application and belongs to the field of genetic engineering. The method comprises the steps of firstly, extracting genomic DNA from a chicken species to be detected, then, amplifying 752bp fragments of upstream and downstream of a locus rs16492031 in an intron 32 of the gene GARNL1, then, subtyping an obtained PCR product by using RFLP, and then, carrying out statistical analysis on correlation between different genotypes of the locus rs16492031 of the gene GARNL1 andthe cockscomb and wattle traits of the cock. According to the application, a relationship for detecting correlation between polymorphism of the locus rs16492031 in the intron of the gene GARNL1 of thechicken species and the cockscomb and wattle traits of the cock is employed, and this relationship and the method serve as means for breeding and marking chicken species. Compared with the present generation breeding, the breeding method has the advantages that the cost is low, the operation is simple and convenient, and the result is accurate.
