Embodiment Construction

[0028] The present invention is explained in detail below in conjunction with examples.

[0029] 1. Primer design and screening

[0030] Based on the design of fluorescent quantitative PCR amplification primers for identification of goat poxvirus by high-resolution melting curve (HRM), 34 full-length genome sequences of goatpoxvirus were downloaded from NCBI, of which 10 full-length genome sequences of GTPV Long genome sequences, 9 full-length genome sequences of SPPV, 15 full-length genome sequences of LSDV. These sequences were compared by multiple sequences using Megalign to find the segments with common conserved sequences at both ends of the GTPV, SPPV, and LSDV genomes and obvious differences in the middle sequences, and primer3.0 was used to design common primers for this segment. The primers can simultaneously amplify target fragments in GTPV, SPPV, and LSDV, but the amplified products can represent different viruses respectively. Through the analysis of 34 whole gen...

Abstract

The invention relates to a virus detection kit and a use method thereof, in particular to a kit for detecting and identifying capripoxvirus (goatpox virus, sheeppox virus and lumpy skin disease virus)and a use method for non-disease diagnosis. The kit for detecting and identifying the capripoxvirus (goatpox virus, sheeppox virus and lumpy skin disease virus) comprises specific primers of which the sequences are SEQ ID No. 1 and SEQ ID No. 2. The kit has high specificity and high sensitivity. The kit is high in cost performance, can screen unknown mutations on any amplicon by only needing a pair of specific primers, and is greatly reduced in cost and simple and convenient to operate compared with a Taq-man probe method.

Description

technical field [0001] The invention relates to a virus detection kit and its use method, in particular to a kit and its use method for detection and identification of goatpox virus (goat pox virus, sheep pox virus, bovine nodular skin disease virus) . Background technique [0002] Capripoxvirus (CPV) belongs to the Poxviridae Vertebrate Poxvirus Subfamily (Chordopoxvirinae), including goatpoxvirus (Goatpox Virus, GTPV), sheep poxvirus (SheeppoxVirus, SPPV) and bovine nodular skin disease virus (Lumpy Skin Disease Virus, LSDV) three members. Goatpox virus members can cause bovine nodular skin disease (Lumpy skin disease), goat pox (Goatpox) and sheep pox (Sheeppox) and other animal diseases. Bovine nodular dermatosis, also known as bovine nodular skin disease or bovine nodular rash, clinically manifests as extensive nodules on the skin, mucous membranes and internal organs of sick cattle, enlarged lymph nodes, skin edema, and local formation of hard nodules or Blight char...
