Embodiments of the present device include a flow regulating pathway having a hydrophobic region in contact with a capillary pathway comprising at least one stepped-down junction. In some embodiments, the device consists of two substrates in contact with each other and a capillary passage between the substrates. In some embodiments, the capillary pathway consists of grooves in one or more substrates. In some embodiments, the capillary passage consists of a spacer present between the two substrates.
