Abstract

The invention relates to a method for culturing and obtaining the monoploid plant of Zantedeschia aethiopica with free microspores, belonging to the technical field of flower breeding. The steps of the method comprise that the Zantedeschia aethiopica stamen with the microspore being at the late nonnuclear stage is fetched for sterilization; the microspore is free mechanically and the cell densityis adjusted to 1.0*10<5> microspores / ml with NLN-13% fructose and then the free microspores are pre-cultured in the dark under the temperature of 32 DEG C for two days; and then the microspores are pre-cultured in the dark under the temperature of 25 DEG C for five days and the culture medium is changed for the culture medium of NLN-13% sucrose after culture; continuously the microspores are cultured in the dark under the temperature of 25 DEG C and the microspores are developed into callus; the callus forms the monoploid plant by bud differentiation. The monoploid plant of Zantedeschia aethiopica obtained by the method needs only one generation to obtain the pure strain with stable heredity by doubling the chromosome, which is an approach for accelerating the purification of breeding materials and process effectively.
