[0034] The method for preparing an antigen vaccine according to the present invention has the following advantages:

[0035] - have all the advantages of an autologous vaccine, i.e. its specificity with respect to a particular patient;

[0036] - have all the advantages of multivalent vaccines, i.e. antigenic diversity;

[0037] - Vaccine enrichment of surface antigens of endothelial cells (preferably activated endothelial cells) after immunization can overcome immune tolerance to tumor vascular endothelial cells brought about by immunization;

[0038] - The method allows obtaining antigens sufficient for vaccination doses by accumulation of antigens rather than proliferation of endothelial cells. This is important because primary cell cultures gradually change their antigenic structure during in vitro proliferation, rendering them unusable for vaccination.

[0039] The practical application of the surface antigen obtained according to the present invention depends on the ch...

Abstract

The inventive method for producing an antitumoral vaccine using endothelial cells, consists in exposing live endothelial cells to a protease action which is vital therefor and in extracting the released surface antigens. The live endothelial cells are repeatedly treated with protease at intervals necessary for restoring the surface antigens by the cells, the surface antigens are accumulated untilthe dose thereof, necessary for vaccination, is obtained and the quality of the thus obtained vaccine is verified. Said invention makes it possible to increase the efficiency of oncological disease treatment by damaging tumoral vessels by overpassing the immune tolerance of the body to the endothelial cells of the tumoral vessels. The invention keeps in mind the overpassing of immune tolerance, inparticular to activated endothelial cells, thereby making it possible to damage the immune system, in particular of tumoral vessels.

Description

technical field [0001] The invention relates to the field of medical technology, in particular to the preparation of tumor immune vaccines, which can be applied to the treatment of tumors and the medicines for preventing their recurrence. Background technique [0002] The limitations of surgical methods and chemotherapy on tumor treatment make further development of new treatment methods for tumor patients urgent. Special attention is now being paid to anti-angiogenic therapies, the principle of which is based on inhibiting the growth of tumor blood vessels. [0003] There are two main stages of tumor development: prevascular and vascular. In the first stage, tumors grow by obtaining nutrients and oxygen from the patient's blood vessels by diffusion, which confines tumor growth to less than a few cubic millimeters. Next, the growth of the tumor needs to transition to the second stage, which is tumor vascularization (Folkman J., "What is evidence that tumors are angiogenesi...
