The high-accuracy vertical axis inclination angle measurement system comprises a vehicle-mounted platform, a photoelectric theodolite, a television auto-collimator and a plane reflecting device, wherein the photoelectric theodolite is fixedly arranged on the vehicle-mounted platform and carries a first imaging device; the television auto-collimator carries a second imaging device; the plane reflecting device is positioned right below the television auto-collimator and is fixed relative to the ground; the television auto-collimator is vertically fixed below a base of the photoelectric theodolite. The calibration method comprises the following steps: coarsely leveling the photoelectric theodolite; starting the television auto-collimator; arranging a collimator which is horizontally placed at the same horizontal position of the photoelectric theodolite; inclining the vehicle-mounted platform; rotating an azimuth axis of the photoelectric theodolite; and re-inclining the vehicle-mounted platform.
