Embodiment Construction

[0062] We developed specific antibodies against mycobacterial species (PPD, BCG and MTB) in vitro. The antibody is immobilized on a biosensor surface plasmon resonance (SPR) system, which can identify particles in serum / plasma of TB patients with high sensitivity and specificity.

[0063] The findings could be used as a basis for developing diagnostic tests to

[0064] - Diagnosis of TB infection within the patient

[0065] - Distinguish between different M. tuberculosis infections (M. tuberculosis, M. bovis, non-tuberculosis mycobacterium infections, etc.)

Abstract

The present invention relates to a method for determining whether an individual has an active tuberculosis infection, a latent tuberculosis infection or is vaccinated against a tuberculosis infection or being healthy. The method comprises contacting a body fluid sample from an individual with an antibody having specificity for a Mycobacterium tuberculosis (Mtb) protein mimicking a human protein and studying the interaction between the protein in the sample and the antibody. High levels of proteins interacting with the antibody are indicative of an active Mtb infection and low levels of proteins interacting with the antibody are indicative of a latent Mtb infection. No levels of proteins interacting with the antibody are indicative of a vaccinated or healthy individual. The present invention also relates to a method determining whether the individual has a tuberculosis infection and to a method for detecting the potency of a vaccination against tuberculosis by contacting a sample from the individual with an antibody having a specificity for the Mtb or the BCG vaccine respectively and studying the interaction between the sample and the specific antibody.

Description

technical field [0001] The present invention relates to methods of detecting tuberculosis infection in an individual and methods of detecting whether an individual has been vaccinated against tuberculosis. Background of the invention [0002] Tuberculosis (TB) is an infectious disease caused by Mycobacterium tuberculosis (Mtb) 1,2 . It is currently one of the leading causes of death from infectious diseases worldwide and thus poses a major public health problem 2 . Although the number of TB cases has declined since 2006, an estimated 3 million deaths are reported each year among 8-10 million infected people 1,3 . About 2 billion people are latently infected with Mtb, and according to the WHO, it causes 9.8 million new cases and about 1.8 million deaths a year worldwide 1,3 . The problem is compounded with the emergence of multidrug-resistant (MDR) and extensively drug-resistant (XDR) strains 2,4 . The most common method used worldwide to diagnose TB is sputum smear m...
