The invention discloses a positioning method and a positioning device for a datum point in a medical image. The positioning method comprises the steps of: forming a first region of the medical image based on a first threshold value of image intensity; forming a second region of the medical image based on a second threshold value of image intensity, wherein the second threshold value is less than the first threshold value; determining seed points in the first region; and screening the second region covering the seed points to obtain the datum point. The positioning method and the positioning device are used for carrying out automatic positioning processing on the datum point in the medical image, save operation time for doctors, and avoid errors of manual positioning. Furthermore, the positioning method and the positioning device are fast in datum point positioning speed and high in datum point positioning accuracy, and are suitable for recognizing and positioning datum points of various types.
