Abstract

The invention discloses a high-efficiency method for ligating with adapters. The method comprises the following steps of using A type DNA polymerase or DNA polymerase Klenow segments to process DNA molecules so as to add a 3'dA protruding terminus; then ligating the processed DNA molecule with an adapter mixture to obtain a ligating product, wherein the adapter mixture comprises three kinds of double-strand DNA adapters; certain ends of the three double-strand DNA adapters are a blunt end, a 3'dT protruding terminus and a 3'dC protruding terminus. According to the method, the ligating efficiency of the adapters and the DNA can be effectively improved.
