Promotion of healing of intestinal mucosa using proline, serine and threonine

A technology of proline and serine, which is applied in the field of health and nutrition, can solve the problems that the role of dietary supplements has not been fully studied, and there is no treatment for IBD patients with mucosal healing, so as to restore intestinal barrier function, repair intestinal epithelial cells, and promote The effect of protection

Problems solved by technology

Although epithelial intima and barrier repair are now recognized to be important for achieving effective IBD therapy, there are no effective treatments to promote mucosal healing in IBD patients
Furthermore, in this regard, mucosal healing reduces the risk of relapse in patients with inflammatory bowel disease, but the role of dietary supplements in this process is understudied

Embodiment 1

[0064] Experimental procedures were performed in accordance with the European Guide for the Care and Use of Laboratory Animals (Directive 2010 / 63 / UE). Thirty male Sprague-Dawley rats from Janvier (France), aged 6-8 months and weighing approximately 500 / 700 g on the day of arrival, were used in this study. Animals were housed individually in cages. During the study, they had free access to food and drinking water or dextran sodium sulfate (DSS, MW 36-44 kDa, ICN Biomedicals) solution ad libitum. Colitis was induced by treating rats with 5% DSS from D0 to D8 (acute colitis) followed by 2% DSS from D9 to D28 (chronic colitis). DSS was dissolved in autoclaved water and given to rats ad libitum. Control animals were provided with water that did not contain DSS from D0 to D28.

[0065] 3 groups of rats received the following treatments and diets :

[0066] The CTRL group (CTRL-ALA, n=10) received a control diet which was a dry semi-synthetic powder (g / kg) consisting of: carboh...

Abstract

Methods and compositions are provided that use proline, serine and threonine to promote healing of the intestinal mucosa. A therapeutically effective amount of proline, serine and threonine is administered to an individual in need thereof, for example a human or other mammal, such as an individual that has damaged intestinal mucosa. The methods and compositions can promote mucosal healing, restore gut barrier function, repair intestinal epithelial cells, promote protection of the colonic epithelium, and / or promote the replenishment of goblet cells in the intestinal and colonic mucosa.

Description

Background technique [0001] This disclosure generally relates to health and nutrition. More specifically, the present disclosure relates to methods and compositions for promoting intestinal mucosal healing using proline, serine, and threonine. [0002] The mucosa is the innermost layer of the gastrointestinal tract and surrounds the lumen passing through the gastrointestinal tract. The gastrointestinal mucosa is formed by the innermost layer, the epithelium; a thin layer of smooth muscle surrounding the skin, the muscular mucosa; and a layer of connective tissue between the epithelium and the muscular mucosa, the lamina propria. [0003] Inflammatory bowel diseases (IBDs), including ulcerative colitis and Crohn's disease, are characterized not only by mucosal inflammation but also by severe impairment of intestinal barrier function. Recent clinical studies have characterized "mucosal healing" as the most significant prognostic factor for long-term remission in IBD patients a...
